Abstract
In Quang Tho commune, Quang Dien district, Thua Thien Hue province, Centella is a primary product to increasing farmers’ income. To provide disease-free plantlets for organic cultivation, we developed a micropropagation method for this culture. The stem explants washed and sterilized with 0.1% HgCl2 for 8 min exhibit the highest survival rate of 66.67%. The SH salt medium supplemented with 30 g/L sucrose, 9 g/L agar, and 1 mg/L BAP is suitable for culturing stem segments, with 3.37 regenerated shoots per sample. The medium for shoot multiplication is SH supplemented with 2 mg/L BAP, 0.2 mg/L TDZ, and 1 mg/L KIN, providing 7.7 shoots per sample. The roots form and develop well on the ½ SH medium supplemented with 1 mg/L IBA, with 14.83 roots per shoot. The suitable substrate for in vitro plant growth is a mixture of 50% soil and 50% rice husk, with a 92.24% survival rate.
