Abstract
This study aimed to establish culture conditions for peripheral blood mononuclear cells (PBMCs) from dogs (Canis lupus familiaris) for application in immunological models and in vitro testing. PBMCs were isolated from whole blood using Ficoll gradient centrifugation and then cultured in 96-well plates using two different media (RPMI-1640 and DMEM) supplemented with three concentrations of fetal bovine serum (FBS: 5%, 10%, 15%). Parameters including cell morphology, cell density, viability of suspended cells, and adhesion cell viability (assessed by the MTT assay) were evaluated at days 1, 3, 6, and 9. The results showed that RPMI-1640 medium maintained a higher density of suspended cells, particularly on day 6 (2.733 ± 0.104 ×10⁵ cells/mL; p < 0.05), while DMEM supplemented with 10% FBS promoted better cell adhesion and sustained higher MTT optical density (OD) values, reflecting the presence of more viable cells with active intracellular metabolic enzyme activity. There was no significant difference in viability or MTT OD between the 10% and 15% FBS groups, indicating that 10% FBS is an appropriate concentration for culture applications.
